What a pharmaceutical cleanroom means under GMP

In the UK, a pharmaceutical cleanroom is graded under EU GMP, which the MHRA enforces through the Orange Guide. There are four grades, A to D. Each is defined by a particle limit measured in two states: at rest, and in operation with people working.

The grades map onto ISO 14644-1 classes. Grade A and B run at ISO 5 at rest. Grade C is ISO 7 at rest and ISO 8 in operation. Grade D is ISO 8 at rest. So most non-sterile production, solution prep, and support areas sit at Grade C or D, which is the ISO 7 and ISO 8 envelope a Monmouth modular cleanroom is built to deliver.

The process sets the grade you need. Terminally sterilised products and component prep run at Grade C and D. Aseptic processing needs a Grade A critical zone inside a Grade B background. Get the grade wrong and the audit finds it. Where the modular kit fits, and where it doesn't

Validation, Annex 1 and audit readiness

Buying the envelope is the first decision. The harder one is who validates it, monitors it, and stands behind it when an MHRA inspector or a customer auditor walks the floor. Monmouth Scientific provide Installation Qualification and Operational Qualification documentation, particle count testing to ISO 14644-1, HEPA integrity testing, pressure cascade data, and the design specification pack. This is the evidence a Qualified Person asks for. Performance Qualification is process-specific and runs once the suite is handed over.

GMP Annex 1 was revised in 2022 and took effect in 2023, with a sharper focus on the Contamination Control Strategy and continuous monitoring. Build for it now and you avoid a retrofit later.
